Erythromycin inhibits Cl secretion

چکیده
We read with interest the report of erythromycin reducing chloride secretion in canine tracheal epithelium (1]. However, we would like to caution against extrapolating from dog to man. Canine trachea is virtually unique in its ion transport properties predominantly secreting chloride, a process which is stimulated by endogenous prostaglandin synthesis. The shortcircuit current of human airways principally relates to absorption of sodium and chloride secretion represents only a small component of this current [2]. Furthermore, it is presently unknown whether endogenous prostaglandins have any role in influencing these processes. Thus, similar studies in a sodium absorbing epithelium will be needed to assess the likelihood of the hypothesis that erythromycin alters ion and hence water transport in man. Finally, at present there is no direct evidence that we know of which demonstrates that altering airway ion transport affects the depth of the sol phase of airway surface fluid and the rheological properties of mucus to alter mucociliary clearance. Studies with amiloride [3, 4 J are suggestive that there may be such effects, but these tenets have yet to be rigorously explored. E.W.F.W. AI.ton, Lecturer in Thoracic Medicine, D.M. Erythromycin inhibits Cl secretion across canine tracheal epithelial cells.
We studied the effect of the macrolide antibiotic erythromycin on bioelectrical properties of canine cultured tracheal epithelium under short-circuit conditions in vitro. Addition of erythromycin to the submucosal but not to the mucosal side dose-dependently decreased short-circuit current (Isc), the maximal decrease from the baseline value and the concentration required to produce a half-maxim...
متن کاملEstrogen inhibits chloride secretion caused by cholera and Escherichia coli enterotoxins in female rat distal colon.
Excessive Cl(-) secretion is the driving force for secretory diarrhea. 17β-Estradiol has been shown to inhibit Cl(-) secretion in rat distal colon through a nongenomic pathway. We examined whether 17β-estradiol inhibits Cl(-) secretion in an animal model of secretory diarrhea and the downstream effectors involved. The effect of 17β-estradiol on cholera toxin and heat-stable enterotoxin induced ...
متن کاملTolvaptan inhibits ERK-dependent cell proliferation, Cl⁻ secretion, and in vitro cyst growth of human ADPKD cells stimulated by vasopressin.
In autosomal dominant polycystic kidney disease (ADPKD), arginine vasopressin (AVP) accelerates cyst growth by stimulating cAMP-dependent ERK activity and epithelial cell proliferation and by promoting Cl(-)-dependent fluid secretion. Tolvaptan, a V2 receptor antagonist, inhibits the renal effects of AVP and slows cyst growth in PKD animals. Here, we determined the effect of graded concentratio...
متن کاملThe antifungal antibiotic, clotrimazole, inhibits Cl- secretion by polarized monolayers of human colonic epithelial cells.
Clotrimazole (CLT) prevents dehydration of the human HbSS red cell through inhibition of Ca++-dependent (Gardos) K+ channels in vitro (1993. J. Clin Invest. 92:520-526.) and in patients (1996. J. Clin Invest. 97:1227-1234.). Basolateral membrane K+ channels of intestinal crypt epithelial cells also participate in secretagogue-stimulated Cl- secretion. We examined the ability of CLT to block int...
متن کاملAntibacterial agents and release of periplasmic pertussis toxin from Bordetella pertussis.
Pertussis toxin accumulates in the periplasm of Bordetella pertussis prior to secretion, and we examined its fate following treatment with antimicrobial agents. Both antibiotics that inhibit protein synthesis (erythromycin and chloramphenicol), transcription (rifampin), or cell wall biosynthesis (cefoperazone and piperacillin) and magnesium sulfate (which inhibits transcription of pertussis tox...
